Lambda Square Mapping & FLIM

Leica Microsystems has developed Lambda Square Mapping (λ2) and FLIM for the comprehensive and easy characterization of complex fluorescent sample. Both systems are based on the white light laser confocal system Leica TCS SP5 X. λ2 identifies each fluorescent molecules by their characteristic excitation- emission spectrum which is acquired with the tunable white light laser. Using this information, imaging settings can be optimized for the highest image contrast and best data quality.

The FLIM method uses the pulsed white light laser as continuous tunable excitation source for the investigation of the characteristic properties of fluorophores. Additionally, it provides information about the direct environment of a molecule, like local pH or binding to another molecule.
